This peptide corresponds to 15 amino acids near the carboxy terminus of human Mcl-1.
PEP-0238 can be used as a blocking peptide with polyclonal antibody PA5-20120.

Target Information

MCL1 (Myeloid cell leukemia-1) belongs to the Bcl-2 family and is involved in programming, differentiation, and maintenance of cell viability, but not proliferation. Isoform 1 of MCL1 inhibits apoptosis, while isoform 2 promotes it. The carboxy terminus of MCL1 and Bcl-2 share significant sequence homology.

Expression of MCL1 increases upon exposure of ML-1 cells to DNA-damaging agents (e.g., ionizing radiation, ultraviolet radiation, alkylating drugs), accompanied by increases in GADD45 and Bax and a decrease in Bcl-2. Enhanced MCL1 expression, associated with mitochondria, complements Bcl-2 expression in differentiating ML-1 cells.

Like Bcl-2, MCL1 promotes cell viability under apoptotic conditions. Although the exact mechanism of apoptosis inhibition is unknown, it likely involves heterodimerization with pro-apoptotic Bcl-2 family members such as Bim or Bak. MCL1 is expressed in multiple cell types and is essential for embryogenesis and lymphocyte development. Two isoforms, MCL1L and MCL1S, exist, with MCL1S promoting cell death when overexpressed.
