ErbB4 (Her-4, c-erbB-4, v-erbB-4)는 class I receptor kinase family의 구성원으로, 약 500개 이상의 유전자 산물을 포함하는 단백질 키나아제 패밀리 중 하나입니다.
이 패밀리는 PTK(tyrosine kinase) 또는 STK(serine/threonine kinase) 촉매 도메인의 서열 비교를 기반으로 8개의 주요 그룹으로 분류됩니다.
ErbB4는 neuregulins 및 NTAK(neural and thymus derived activator for ErbB kinases)에 의해 활성화되며, heregulin의 수용체로 작용하여 HGL 자극에 의한 자가 인산화를 매개합니다.
이 단백질은 세포 대사, 전사, 세포주기 진행, 세포 골격 재배열, 세포 이동, 세포사멸, 분화 등 다양한 세포 신호 전달 경로를 조절합니다.
ErbB4는 정상 골격근, 심장, 뇌하수체, 뇌, 소뇌 등에서 발현되며, 특정 유방암 세포주(T47-D, MDA-MB-453, BT-474, H3396 등)에서 높은 발현을 보입니다.
일부 암 세포주에서 ErbB4 수준이 높게 나타나며, 이는 특정 종양의 발생과 관련이 있을 수 있습니다.
