Target Information

RUVBL2 possesses single-stranded DNA-stimulated ATPase and ATP-dependent DNA helicase (5′ to 3′) activity. It is a component of the NuA4 histone acetyltransferase complex involved in transcriptional activation of select genes through histone acetylation (H4 and H2A). This modification alters nucleosome–DNA interactions and promotes recruitment of transcription-regulating proteins. The NuA4 complex is implicated in oncogene- and proto-oncogene-mediated growth induction, tumor suppressor-mediated growth arrest, replicative senescence, apoptosis, and DNA repair. RUVBL2, together with RUVBL1 and EP400, contributes to the ATPase and helicase activities of NuA4 and plays a direct role in DNA repair. It is essential for MYC-mediated oncogenic transformation and modulates transcriptional activation by the LEF1/TCF1-CTNNB1 complex. RUVBL2 may also inhibit the transcriptional activity of ATF2.
