Target Information

This gene encodes a G protein-coupled receptor with selectivity for three chemokines: IP10 (interferon-γ-inducible 10 kDa protein), Mig (monokine induced by interferon-γ), and I-TAC (interferon-inducible T cell α-chemoattractant). These chemokines belong to the CXC subfamily, characterized by a single amino acid separating the first two conserved cysteine residues.
Binding of chemokines to CXCR3 induces cellular responses involved in leukocyte trafficking, including integrin activation, cytoskeletal changes, and chemotactic migration. Inhibition by Bordetella pertussis toxin suggests coupling with Gi-class heterotrimeric G proteins.
Phosphorylation-dependent receptor internalization leads to rapid, transient responses, followed by receptor recycling upon dephosphorylation. CXCR3 is highly expressed in effector/memory T cells and inflamed tissues, where it mediates recruitment of inflammatory cells.
This receptor is a potential target for small-molecule antagonists for treating inflammatory diseases. Multiple transcript variants encoding different isoforms have been identified.
