Target Information

The MDM2 protein is the primary regulator of p53 protein stability. MDMX (also known as MDM4) is an MDM2-related protein that inhibits MDM2-mediated degradation of p53 through distinct interactions. The MDMX gene is often amplified in malignant gliomas. ARF binds and sequesters MDMX within the nucleolus, leading to increased p53 transactivation. Expression of MDMX can reverse MDM2-mediated p53 degradation while maintaining suppression of p53 transactivation. MDMX also binds and stabilizes p73, increasing its half-life in contrast to p53.
