Target Information
Beta-arrestin-2 (ARRB2) is an adaptor protein in the arrestin family with N- and C-arrestin domains. It regulates G protein-coupled receptor desensitization, particularly beta2-adrenergic receptor (beta2AR) function, by enhancing beta2AR-mediated nuclear translocation of ERK. ARRB2 binds phosphorylated beta2ARs, impairing their G(S) protein activation capacity. Together with AIP4, ARRB2 mediates CXCR4 endosomal sorting into a degradative pathway and may participate in hormone-specific desensitization of TSH receptors. It acts as a Ca²⁺ binding protein in retinal outer segments, interacts with P-rhodopsin, and contributes to synaptic transmission in photoreceptor cells. ARRB2 is widely expressed, notably in neuronal tissues and spleen.
