Target Information
Focal Adhesion Kinase (FAK)는 125 kDa의 비수용체형 단백질 티로신 키나아제로, Src의 기질로 작용하며 인테그린 신호전달의 핵심 요소입니다. FAK는 세포 부착, 분화, 이동, 세포사 및 세포주기 G1에서 S 단계로의 전이에 중요한 역할을 합니다.
FAK는 중심 촉매 도메인과 C-말단 꼬리를 가지며, 이는 세포가 세포외기질에 부착하는 초점부착(focal adhesion) 부위로의 위치를 결정합니다. 인테그린이 피브로넥틴과 결합하면 FAK의 티로신 인산화가 증가하며, Grb2 및 c-Src 단백질과의 결합을 유도하여 ERK2 MAP kinase를 활성화합니다.
Tyr925 부위는 Grb2 SH2 도메인과 직접 결합하는 부위로 확인되었으며, 이는 세포 부착 의존적 신호전달 조절에 중요한 역할을 합니다.
