Target Information

CD20 is a non-glycosylated surface phosphoprotein with a molecular weight range of 33–37 kDa depending on phosphorylation. It is expressed on mature and most malignant B cells, a subpopulation of T lymphocytes, and follicular dendritic cells. CD20 expression coincides with surface IgM and regulates calcium conductance, cell cycle progression, and B-cell proliferation.

CD20 is associated with lipid rafts, with association intensity influenced by extracellular triggers, conformational changes, and BCR aggregation. After receptor ligation, BCR and CD20 colocalize and then dissociate before BCR endocytosis, while CD20 remains on the cell surface. CD20 is a useful target for antibody-mediated B-cell depletion therapy, as it is highly expressed on most B-cell malignancies and remains membrane-bound following antibody treatment.

Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
