• Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at room temperature for 20 minutes before staining.

Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into the cytoplasm of T cells and regulating TCR complex expression.

• CD3 subunits (gamma, delta, epsilon, zeta) are immunoglobulin superfamily members encoded by genes on human chromosome 11.
• The cytoplasmic tails of CD3 subunits associate with signal transduction molecules via a double tyrosine-based motif.
• CD3 is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum.
• Not expressed on B or NK cells.
• CD3 defects are associated with CD3 immunodeficiency and may affect T lymphocyte percentages in autoimmune diseases.
