Target Information

STK3 (MST2) is a STE kinase family member that acts upstream of the MAP kinase signaling pathways and is activated by proapoptotic stimuli such as staurosporine and FAS ligand.
It functions as a stress-activated, pro-apoptotic kinase that, following caspase cleavage, enters the nucleus to induce chromatin condensation and DNA fragmentation.
STK3/MST2 is a key component of the Hippo signaling pathway, regulating organ size and tumor suppression by restricting proliferation and promoting apoptosis.
In this pathway, STK3/MST2 and STK4/MST1, in complex with SAV1, phosphorylate and activate LATS1/2 with MOB1, which subsequently phosphorylates and inactivates YAP1 and WWTR1/TAZ.
Phosphorylation of YAP1 by LATS2 inhibits its nuclear translocation, controlling genes related to proliferation, apoptosis, and migration.
STK3/MST2 and STK4/MST1 are essential for repressing hepatocyte proliferation, preventing activation of adult liver stem cells, and inhibiting tumor formation.
