Target Information
The p38 MAPK cascade regulates various cellular responses to stress, inflammation, and other stimuli. p38 MAPK is inactive in its nonphosphorylated form and becomes rapidly activated by dual phosphorylation of Thr-Gly-Tyr motifs. There are four isoforms of p38 MAPK, differing in tissue expression and affinity for activators and effectors. Activation occurs upon exposure to TNF, IL-1, heat shock, or other stimuli, leading to phosphorylation by MAPK kinase-3/6 at Thr180 and Tyr182. Activated phospho-p38 MAPK phosphorylates transcription factors such as ATF-2, CHOP-1, and MEF-2.
