HERC5 (E3 ISG15-protein ligase HERC5)는 ISG15 conjugation의 주요 E3 ligase로, interferon에 의해 유도된 선천적 항바이러스 반응의 양성 조절자로 작용합니다.
HERC5는 ISGylation machinery의 일부로서, 넓고 비특이적인 방식으로 표적 단백질을 인식합니다. IRF3의 ISGylation을 촉진하여 지속적인 활성화를 유도하고, IRF3-PIN1 상호작용을 억제하여 IRF3의 유비퀴틴화 및 분해를 방지함으로써 항바이러스 반응을 강화합니다.
또한 HERC5는 폴리리보솜과 물리적으로 결합되어 새로 합성되는 단백질을 공번역적으로 수정합니다.
