Target Information
BACE1 (beta-secretase)는 Alzheimer’s disease(AD)에서 발견되는 amyloid beta-peptides(Abeta) 생성에 관여하는 주요 효소입니다. 일부 조기 발병형 AD는 amyloid precursor protein(APP)의 ‘Swedish’ 변이에 의해 BACE1 절단이 증가하면서 발생합니다. 이러한 유전적 및 병리학적 근거로 BACE1과 gamma-secretase 억제가 치료 접근법의 주요 목표가 되고 있습니다.
APP는 먼저 beta-secretase에 의해 절단되어 용해성 단백질 파생물과 막에 고정된 99-아미노산 C-말단 조각(C99)을 생성합니다. 이후 C99는 gamma-secretase의 기질로 작용하여 4 kDa amyloid-beta 펩타이드를 생성하며, 이는 AD 환자의 뇌에 침착됩니다. BACE1은 beta-site APP cleaving enzyme(BACE) 또는 aspartyl protease 2(Asp2)로도 알려져 있으며, APP와 공위치합니다.
