Target Information
The expression pattern of the Prox1 homeobox gene suggests a role in various embryonic tissues, including the lens. Analysis of mRNA reveals that Prox1 mRNA is present in many human tissues, with the highest level in the lens. Homozygous Prox1⁻/⁻ mice die at midgestation from multiple developmental defects, including lens abnormalities. Prox1 inactivation causes abnormal proliferation, downregulation of Cdkn1b and Cdkn1c, misexpression of E-cadherin, and excessive apoptosis, leading to defective lens formation.
Prox1 is expressed in endothelial cell subpopulations that form the lymphatic system and is a key regulator of lymphatic development. It is also required for hepatocyte migration in mice; loss of Prox1 results in smaller livers with reduced hepatocyte clusters. In the embryonic mouse retina, Prox1 regulates progenitor cell cycle exit and differentiation, influencing proliferation and cell-fate determination.
