Target Information
Estrogen Receptor beta (ERβ, NR3A2) is approximately 55 kDa, with multiple isoforms ranging from 36–59 kDa. Initially cloned from human testis, ERβ belongs to the steroid/thyroid hormone receptor superfamily of nuclear receptors.
It acts as a ligand-activated transcription factor that binds estrogen, dimerizes, and interacts with estrogen response elements (EREs) in DNA to regulate gene transcription.
Two forms exist—ERα and ERβ—encoded by ESR1 and ESR2, respectively. ERβ binds estrogens with similar affinity to ERα and can inhibit ERα activity in reproductive tissues.
ERβ is expressed in various tissues and functions as a tumor suppressor, implicated in prostate cancer, autism spectrum disorder, medullary thyroid carcinoma, De Quervain disease, and cardiovascular disease risk.
