Target Information
Anaplastic Lymphoma Receptor Tyrosine Kinase (ALK) belongs to the insulin receptor superfamily and is essential for brain development. Mutations, rearrangements, and amplifications in the ALK gene have been identified in various tumors, including anaplastic large cell lymphoma, neuroblastoma, and non-small cell lung cancer. Chromosomal rearrangement is the most common genetic alteration, often resulting in a fusion gene between ALK and nucleophosmin (NPM). The NPM-ALK fusion product is oncogenic. ALK encodes a receptor protein-tyrosine kinase with transmembrane and extracellular domains, which are absent in the NPM-ALK fusion protein. ALK shares high sequence similarity with LTK and plays a critical role in neuronal development.
