Target Information
Reticulon-1 (RTN1), formerly known as Neuroendocrine-Specific Protein (NSP), is expressed in neural and neuroendocrine tissues. The NSP gene is located on human chromosome 14q21–q22 and encodes three overlapping proteins: reticulon-1A (NSP-A), reticulon-1B (NSP-B), and reticulon-1C (NSP-C).
These proteins are anchored to endoplasmic reticulum membranes via their common C-terminal regions.
- Reticulon-1A: ~135 kDa, multiple isoforms depending on serine phosphorylation; serves as a marker for neuroendocrine differentiation in small cell lung carcinoma (SCLC) and carcinoid tumors.
- Reticulon-1B: 45 kDa phosphoprotein, found only in lung cancer cell line NCI-H82, not detected in normal tissues.
- Reticulon-1C: 23 kDa non-phosphorylated protein, co-expressed with Reticulon-1A in SCLC but absent in non-SCLC cultures.
