Target Information
Mitochondrial transcription factor A (TFAM) is a nuclear-encoded protein that is transported to mitochondria, where it plays a key role in maintaining and expressing mitochondrial DNA (mtDNA). TFAM regulates mtDNA replication and transcription, ensuring proper mitochondrial function. It contains an LC3 interacting region (LIR) motif that binds autophagy proteins such as LC3, aiding in removal of leaked mtDNA and limiting inflammation. TFAM levels vary by tissue, influencing oxidative phosphorylation (OXPHOS) capacity and mitochondrial genome stability. Disruptions in TFAM are associated with neurodegeneration and diseases related to mtDNA stress.
