Target Information

Interleukin 8 (IL-8, CXCL8) is a 72 amino acid pro-inflammatory factor belonging to the CXC subfamily of chemokines. It binds to cell surface receptors IL-8RA and IL-8RB and functions as a chemoattractant and potent angiogenic factor.
IL-8 expression and secretion are induced by diverse inflammatory stimuli in macrophages and endothelial cells. In endothelial cells, IL-8 is stored in Weibel-Palade bodies.
Originally isolated from osteosarcoma cells, IL-8 contains the ELR motif (N-terminal Glu-Leu-Arg sequence) and signals through CXCR1 and CXCR2 receptors.
Previous nomenclature includes:

• Neutrophil activating protein 1 (NAP-1)
• Granulocyte chemotactic protein 1 (GCP-1)
• Monocyte-derived neutrophil-activating peptide (MONAP)
• Protein 3-10C

IL-8 and other CXC chemokines form a gene cluster mapped to chromosome 4q.
Cancer studies indicate IL-8’s role in angiogenesis and tumor growth, as well as in the pathogenesis of bronchiolitis, a viral respiratory tract disease.
