Target Information
Ubiquitin-mediated proteolysis involves the transfer of ubiquitin (Ub) to lysine residues on selected cellular proteins, promoting their degradation by the 26S proteasome. Ub-fusions are processed by Ub-specific proteases (UBps) or via the Ub-fusion degradation (UFD) pathway. UFD1L, the human homolog of yeast UFD1, modulates Ub-fusion degradation by selective multiubiquitination. Mutations in UFD1L are associated with CATCH22 syndrome, characterized by cardiac defects, cleft palate, and hypocalcemia, indicating a potential link between proteolytic dysfunction and developmental disorders.
