Target Information

Dynamin is a GTPase required for endocytosis of clathrin-coated vesicles from the membrane. Its role in endocytosis was first identified through the temperature-sensitive shibire mutation in Drosophila melanogaster.
In mammals, three closely related isoforms have been identified:

• Dynamin 1: expressed exclusively in neurons
• Dynamin 2: ubiquitously expressed
• Dynamin 3: primarily expressed in testes

Each isoform has multiple splice variants, suggesting distinct intracellular targets or vesicle budding functions.
Common domain features include:

• Amino-terminal GTPase domain
• Central pleckstrin homology domain
• Proline-rich carboxyl terminus with SH3 domains

Dynamin interacts with proteins such as the AP2 adaptor complex and amphiphysin, targeting it to clathrin-coated pits.
