Target Information
8-hydroxyguanine은 자유 라디칼에 의해 유도되는 산화적 DNA 손상의 한 형태로, G:C → T:A 전환을 초래합니다. E. coli에서는 이러한 손상을 방지하기 위한 세 가지 DNA 복구 효소가 존재하며, 그 중 하나인 MutM은 효모(yOgg1)와 인간(hOgg1) 상동체를 가지고 있습니다.
hOgg1 단백질은 DNA 내에서 피리미딘 반대편의 8-hydroxyguanine을 효율적으로 제거하고, 박테리아 및 효모 효소와 유사한 방식으로 AP 부위를 절단합니다.
손상된 DNA 복구를 조절하는 유전적 배경은 암 발생 민감성과 관련이 있으며, hOgg1 유전자는 다양한 암에서 이형접합성 소실(LOH)이 관찰되는 3p26.2 영역에 위치합니다. 특히, 3p25–p26은 폐암에서 공통적인 LOH 영역입니다.
