Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 activation leads to transcription of genes such as p21waf1, involved in G1 or G2/M arrest.

p53 consists of:

• N-terminal transactivation domain
• Central DNA-binding domain
• Oligomerisation domain
• C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for activation and stabilization, while Ser392 phosphorylation correlates with tumor formation. p53 plays roles in preventing proliferation of damaged cells, aging, oxidative stress, and learning/memory processes.

Mutations in the TP53 gene are common in human cancers, leading to loss of tumor suppressor function. Germline mutations are also found in cancer-prone families such as Li-Fraumeni syndrome.
