Target Information

IRS-1 is a major substrate of the insulin receptor and is phosphorylated in response to stimulation by insulin, insulin-like growth factor 1 (IGF-1), and interleukin 4 (IL-4). It undergoes phosphorylation on serine, threonine, and tyrosine residues in various tissues. Casein kinase II mediates a portion of insulin-stimulated serine/threonine phosphorylation of IRS-1 in vivo, with Thr 502 identified as the major phosphorylation site and Ser 99 as an additional site. IRS-1 contains three putative binding sites for 14-3-3 proteins (Ser 270, Ser 374, and Ser 641), with the motif around Ser 270 located in the phosphotyrosine binding domain responsible for interaction with the insulin receptor.
