Target Information
Tau는 신경세포 축삭에서 주로 발견되는 미세소관 결합 단백질로, 튜불린 중합을 촉진하고 미세소관을 안정화합니다.
C-말단은 축삭 미세소관에 결합하고 N-말단은 신경 세포막 성분에 결합하여 세포 내 연결자(linker) 역할을 합니다.
Tau/MAPT의 위치는 신경세포의 극성을 결정하며, 짧은 아이소폼은 세포골격의 유연성을, 긴 아이소폼은 안정화에 기여합니다.
과인산화된 Tau는 Alzheimer’s disease(AD) 뇌의 paired helical filaments(PHF)의 주요 구성 요소로, 미세소관 결합 기능을 손상시켜 신경세포 퇴화를 유발합니다.
Tau는 GSK-3β, PKA, cdk5, casein kinase II 등 다양한 serine/threonine kinase에 의해 인산화됩니다.
과인산화 Tau는 Pick’s disease, frontotemporal dementia, cortico-basal degeneration, progressive supranuclear palsy 등 여러 신경계 질환에서도 발견됩니다.
