Target Information
Beta-catenin은 세포 간 접착 구조(adherens junction, AJ)의 구성 요소로 처음 확인된 단백질입니다. AJ는 상피 세포층의 형성과 유지에 필수적이며, 세포 성장과 접착을 조절합니다. Beta-catenin은 E-cadherin의 세포질 도메인과 상호작용하여 E-cadherin을 alpha-catenin에 연결하고, 이 복합체는 세포골격의 actin과 결합합니다. 또한 adenomatous polyposis coli 단백질 및 미세소관과 결합하며, tyrosine kinase, phosphatase, Wnt/Wingless 등의 다양한 신호 경로와 상호작용합니다.
Beta-catenin은 모낭 세포, 기저 및 주변 기질 세포, 외부 및 내부 모근초 세포에서 발현되며, pilomatrixoma, medulloblastoma, colorectal cancer, ovarian cancer 등 다양한 종양에서 병리학적 역할이 보고되었습니다. 핵 내에서는 Lef/Tcf 전사인자와 함께 cyclin D, c-myc 등의 유전자 발현을 촉진하여 세포 증식을 유도합니다. 이러한 기능은 여러 고형암(유방, 대장, 간, 폐, 위, 전립선, 피부 등)에서의 종양 발생과 관련이 있습니다.
