Target Information

ICAD (Inhibitor of Caspase-activated DNase) regulates nuclear apoptosis by controlling CAD (Caspase-activated DNase). It exists as two isoforms:

  • ICAD-L/DFF45 (45 kDa): functional isoform
  • ICAD-S/DFF35 (35 kDa): non-functional isoform

ICAD binds and inhibits CAD under non-apoptotic conditions. Upon apoptotic signaling, caspase 3 cleaves ICAD at residues 117 and 224, releasing CAD to fragment nuclear DNA. ICAD also acts as a folding chaperone for CAD. It has been studied as a potential therapeutic target for restoring apoptotic signaling in colon and brain cancers. Two alternatively spliced transcript variants encode distinct isoforms of ICAD.
