Background
The protein encoded by this gene belongs to the SMAD family, which is related to the Drosophila gene mothers against decapentaplegic (Mad) and the C. elegans gene Sma. SMAD proteins act as signal transducers and transcriptional modulators mediating multiple signaling pathways.
This protein mediates bone morphogenetic protein (BMP) signaling, which regulates cell growth, apoptosis, morphogenesis, development, and immune responses. Upon BMP ligand binding, it is phosphorylated by BMP receptor kinase and forms a complex with SMAD4 for transcriptional regulation. It is also a target for SMAD-specific E3 ubiquitin ligases (SMURF1, SMURF2), undergoing ubiquitination and proteasome-mediated degradation.
