Target Information

Ribonucleases (RNAses) H are enzymes that hydrolyze the RNA strands of RNA/DNA hybrids. Their major role is to remove the RNA strand from hybrids formed during DNA replication and repair. RNAse H2 is composed of three subunits, all required for enzymatic activity. Mutations in RNAse H2A or other subunits cause Aicardi-Goutieres syndrome (AGS), a neurological disorder with symptoms similar to viral brain infections, including elevated IFN-alpha levels in cerebrospinal fluid. Similar phenotypes are observed with mutations in TREX1, suggesting a shared role in preventing accumulation of intracellular nucleic acids and activation of innate immune responses.
