Treacle은 1,411 아미노산으로 구성된 serine/alanine-rich 핵 인산화 단백질로, upstream binding factor (UBF)와 상호작용하여 리보솜 DNA 유전자의 전사를 조절합니다. 또한 배아 발생 초기의 두개안면 복합체 발달에 관여하며, 핵소체-세포질 수송에 역할을 할 수 있습니다. Treacle 단백질은 N- 및 C-말단과 큰 중앙 반복 도메인을 포함한 세 개의 도메인을 가지고 있습니다.
TCOF1 유전자의 변이는 Treacher Collins–Franceschetti 증후군(TCS)을 유발하며, 이는 두개안면 발달 결함으로 특징지어집니다. 주요 증상으로는 전도성 청력 손실, 하악 및 상악의 저형성, 하안검 열구의 하향 경사, 구개열 등이 있습니다.
