Product Specific Information
Description:
The UCHT1 monoclonal antibody reacts with human CD3ε, a 20 kDa subunit of the TCR complex. Along with other CD3 subunits (γ, δ), the ε chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Crosslinking of TCR via immobilized UCHT1 initiates intracellular signaling leading to activation and proliferation.
Applications Reported:
Flow cytometric analysis.
Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use 4 µL (0.6 µg) per test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
- Each NovaFluor conjugate or kit ships with CellBlox™ Blocking Buffer.
- Add 5 µL per sample when staining with NovaFluor conjugates.
- For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information
The CD3 subunit complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It includes CD3γ, CD3δ, CD3ε, and CD3ζ subunits, encoded on chromosome 11. These subunits associate with intracellular signaling molecules via tyrosine-based motifs. CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. Altered CD3 expression may be observed in autoimmune diseases. CD3 gene defects are associated with CD3 immunodeficiency.
