Steroid receptors are ligand-dependent intracellular proteins that stimulate transcription of specific genes by binding to DNA sequences following activation by the appropriate hormone. Prior to activation, steroid receptors associate with various proteins, including heat shock proteins (HSP70, HSP90), immunophilins (FKBP52, FKBP54, CyP-40), and other proteins termed p23, p60, and p48.

p23, along with HSP70, HSP90, and p60, forms a transient complex with the progesterone receptor (PR). The cloned human p23 encodes a 160-amino-acid protein highly conserved between species and distinct from previously identified proteins. It is a highly acidic phosphoprotein with an aspartic acid-rich C-terminal domain and multiple phosphorylation sites. In vitro studies indicate that p23 binds to HSP90 and is essential for HSP90 and CyP-40 binding to PR. While its precise function remains unclear, p23 appears to play a critical role in PR activity.
