• Immunohistochemistry (PFA fixed):
  Incubate antibody for 30 minutes at room temperature.
  Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on the surface of mature monocytes, macrophages, and neutrophils. It functions as a multifunctional lipopolysaccharide receptor and exists in both membrane-bound and soluble forms. CD14 binds lipopolysaccharide (LPS) in a reaction catalyzed by LPS-binding protein (LBP), an acute-phase serum protein. The soluble form (sCD14) can differentiate subtle structural variations in LPS and contributes to neutralization of serum lipopolysaccharides by reconstituted lipoprotein particles. CD14 also binds apoptotic cells and influences allergic, inflammatory, and infectious responses. Alternative splicing results in multiple transcript variants encoding the same CD14 isoform. CD14 dysfunction is associated with diseases such as Mycobacterium chelonae infection and croup.
