Target Information

TNF-alpha is a multifunctional proinflammatory cytokine belonging to the tumor necrosis factor (TNF) superfamily. It is mainly secreted by macrophages and binds to its receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR.
TNF-alpha regulates immune cell activity, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It typically exists as multimers (two to five noncovalently linked units) but appears as a single 17 kDa band under non-reducing SDS-PAGE conditions.

Knockout studies in mice suggest TNF-alpha has neuroprotective functions and induces tumor necrosis when injected into tumor-bearing mice. It also plays roles in immune responses to bacterial, viral, parasitic, and fungal infections, and in the necrosis of specific tumors.
TNF-alpha causes cytolysis or cytostasis of certain transformed cells and acts synergistically with interferon-gamma in cytotoxicity. This cytokine is implicated in autoimmune diseases, insulin resistance, and cancer.
