This antibody can recognize both the phosphorylated and unphosphorylated target.

Target Information

RIPK2 is a serine/threonine kinase involved in cell activation, proliferation, differentiation, and apoptosis. Upon stimulation by bacterial peptidoglycans, NOD1 and NOD2 are activated, oligomerize, and recruit RIPK2 through CARD-CARD domains. RIPK2 contributes to tyrosine phosphorylation of ARHGEF2 via Src kinase, leading to NF-kappaB activation by NOD2. Once recruited, RIPK2 autophosphorylates and undergoes Lys-63-linked polyubiquitination by E3 ligases XIAP, BIRC2, and BIRC3. This polyubiquitination promotes recruitment of MAP3K7/TAK1 to IKBKG/NEMO and induces further Lys-63-linked polyubiquitination, activating IKBKB/IKKB. Subsequently, NF-kappaB translocates to the nucleus, activating transcription of genes related to immune response, growth control, and apoptosis protection. RIPK2 also plays a role during T-cell receptor engagement, promoting BCL10 phosphorylation and NF-kappaB activation.
