Target Information

Lysosomal enzymes containing one or two mannose 6-phosphate (man6P) moieties are transported within the cell via approximately 300 kDa cation-independent mannose 6-phosphate receptors (CI-MPR). These receptors participate in two interconnected cycles:

  1. Biosynthetic cycle: Transport of newly synthesized lysosomal enzymes from the trans-Golgi network to late endosomes or early lysosomes.
  2. Endocytic cycle: Transport of extracellular lysosomal enzymes from the plasma membrane via clathrin-coated vesicles to early endosomes.

The entire pool of MPRs cycles between these compartments approximately every 3 hours. The steady-state distribution of MPRs is mainly within late endosomes, fewer in the trans-Golgi network, and about 10% at the cell surface.

In addition to mannose 6-phosphate binding, the MPR also has a binding site for type II insulin-like growth factor (IGF-II) and can bind both ligands simultaneously. A ~240 kDa soluble truncated form representing the extracellular domain of the protein is also found circulating in serum and retains the ability to bind both ligands.
