Target Information
Proteolytic degradation is essential for maintaining proper levels of short-lived and regulatory proteins involved in cellular metabolism, stress response, antigen presentation, receptor modulation, ion channel regulation, cell cycle control, transcription, and signaling.
The ubiquitin-proteasome pathway degrades most cytosolic and nuclear proteins in eukaryotic cells, while others are processed via the vacuolar pathway involving endosomes, lysosomes, and the endoplasmic reticulum.
The 26S proteasome is an ATP-dependent, multisubunit (~31) complex with a molecular weight of ~2.5 MDa, consisting of a 20S core and one or two 19S regulatory subunits. The 19S complex recognizes ubiquitinated proteins and assists in unfolding and translocating them into the 20S core.
Defects in this pathway are associated with various genetic diseases, including cystic fibrosis, Angelman’s syndrome, and Liddle syndrome.
