Target Information
Sphingomyelin synthases synthesize sphingolipids through transfer of a phosphatidyl head group onto the primary hydroxyl of ceramide.
SAMD8 is an endoplasmic reticulum (ER) transferase that lacks sphingomyelin synthase activity but can convert phosphatidylethanolamine (PE) and ceramide to ceramide phosphoethanolamine (CPE) with low product yield.
It operates as a ceramide sensor controlling ceramide homeostasis in the ER rather than converting ceramides. SAMD8 is critical for maintaining the integrity of the early secretory pathway. [UniProt]
