Target Information
LSD1 is a histone demethylase that specifically demethylates Lys-4 of histone H3, a mark associated with epigenetic transcriptional activation, thereby acting as a corepressor. It contains a SWIRM domain, a FAD-binding motif, and an amine oxidase domain. LSD1 functions within histone deacetylase complexes and silences genes by histone demethylation. It oxidizes substrates via FAD to generate imine intermediates that are subsequently hydrolyzed. LSD1 demethylates mono- and tri-methylated Lys-4 of histone H3 and may also demethylate Lys-9, contributing to androgen receptor gene derepression. Mutations in LSD1 can lead to developmental abnormalities including cleft palate and psychomotor retardation.
