This 4D10p35 monoclonal antibody reacts with mouse p35, a component of two members of the interleukin (IL)-12 cytokine family. IL-12 family cytokines are heterodimeric, composed of an alpha-chain (e.g., p35) and a beta-chain (e.g., p40). Expression of the alpha-chain is more restricted, making it the limiting factor for secretion of IL-12 family cytokines. These cytokines are secreted primarily by antigen-presenting cells in response to TLR agonists and other inflammatory stimuli.

The p35 protein is part of IL-12 (IL-12p70), whose secretion is induced by IFN-γ and TLR3, 4, or 7 agonists, and inhibited by Th2 cytokines. IL-12 is essential for Th1 cell differentiation. The p40 subunit is more abundantly expressed and may act independently as an antagonist of IL-12 function.

p35 can also dimerize with EBI3 to form IL-35, an immunoregulatory cytokine produced by regulatory T cells. IL-35 suppresses effector T-cell proliferation and may induce a regulatory phenotype in naive CD4+ cells. Testing of 4D10p35 in the context of IL-35 has not been evaluated.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested by intracellular staining and flow cytometric analysis of stimulated mouse dendritic cells. Recommended ≤0.125 µg/test (100 µL final volume). Optimal titration required.

Target Information

Interleukin 12A (IL-12A) is a signaling cytokine of the IL-6 superfamily. It forms the 35-kDa alpha subunit of IL-12, a disulfide-linked heterodimeric cytokine. IL-12A promotes T and NK cell activation, enhances NK cell lytic activity, and stimulates IFN-γ production. It contributes to Th1/Th2 differentiation and Stat4 phosphorylation. Along with EBI3, IL-12A forms interleukin-35 (IL-35), produced by regulatory T cells for immunosuppressive activity.
