Target Information

HMCES acts as a sensor of abasic sites in single-stranded DNA (ssDNA) and is essential for preserving genome integrity by promoting error-free repair of abasic sites. It recognizes and binds abasic sites in ssDNA at replication forks and forms a covalent cross-link with DNA through a thiazolidine linkage involving its N-terminal catalytic cysteine residue. The HMCES–DNA cross-link is subsequently degraded by the proteasome.

This protein prevents error-prone repair by translesion synthesis polymerases and endonucleases, thereby reducing mutations and double-strand breaks. It shows a preference for ssDNA but can also bind dsDNA with 3′ or 5′ overhangs and dsDNA–ssDNA junctions. Additionally, HMCES participates in class switch recombination (CSR) in B-cells by protecting ssDNA overhangs and promoting DNA double-strand break repair through the microhomology-mediated alternative-end-joining (Alt-EJ) pathway. It also mediates autocatalytic processing of its N-terminal methionine to expose the catalytic cysteine.
