Target Information
Anaplastic Lymphoma Receptor Tyrosine Kinase (ALK) belongs to the insulin receptor superfamily and plays a critical role in brain development. Mutations, rearrangements, and amplifications of the ALK gene have been associated with various tumors, including anaplastic large cell lymphoma, neuroblastoma, and non-small cell lung cancer. The most common alteration is chromosomal rearrangement, generating a fusion gene between ALK and the nucleophosmin (NPM) gene. The NPM-ALK fusion product is oncogenic. ALK encodes a novel receptor tyrosine kinase with transmembrane and extracellular domains, which are absent in the transforming NPM-ALK fusion protein. ALK shows high sequence similarity to LTK and contributes to neuronal development and function.
