Target Information
NANOG (Nanog homeobox) is a divergent homeodomain protein that directs pluripotency and differentiation of undifferentiated embryonic stem cells.
NANOG mRNA is expressed in pluripotent mouse and human cell lines but absent in differentiated cells. Human NANOG shares 52% overall amino acid identity with the mouse protein and 85% within the homeodomain.
Human NANOG maps to gene locus 12p13.31, while mouse NANOG maps to 6 F2. Expression is detected in the inner cell mass of the blastocyst.
Research indicates that high levels of NANOG expression are found in undifferentiated N-Tera embryonal carcinoma cell lines. NANOG functions as a transcription regulator involved in proliferation and self-renewal of embryonic stem cells.
It plays a crucial role in stem cell maintenance and may be useful in generating induced pluripotent stem (iPS) cells for regenerative medicine. Overexpression of NANOG promotes S phase entry and proliferation.
Diseases associated with NANOG dysfunction include teratocarcinoma and germ cell/embryonal cancer.
