Product Specific Information

The 53-2.1 monoclonal antibody reacts with mouse CD90.2 (Thy-1.2), a GPI-linked membrane molecule expressed by thymocytes, mature T cells, and neurons in CD90.2-expressing mouse strains (BALB/c, CBA, C3H, C57BL/6, C58, SJL, etc.). Cells from CD90.1-expressing strains (PL, AKR) do not stain with 53-2.1. CD90 is involved in T-cell adhesion and signal transduction.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.4 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimal titration recommended.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be added to staining cocktails for optimal performance.
  • Add 5 µL per sample (regardless of number of NovaFluors).
  • For single-color controls: add 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD90.2 (Thy-1) is a GPI-linked glycoprotein of the immunoglobulin superfamily. It is expressed in murine T cells, thymocytes, neural cells, Kupffer’s cells, and fibroblasts. It plays roles in cell-cell and cell-ligand interactions during synaptogenesis and other brain processes. In mice, CD90.2 is expressed on all thymocytes and peripheral T lymphocytes, as well as connective tissue and stromal cell lines.
