Product Specific Information

This SDRNR monoclonal antibody recognizes human and mouse AKT (Protein Kinase B, PKB) when phosphorylated on Ser473.
AKT is a serine/threonine kinase involved in metabolism, proliferation, apoptosis/survival, and migration.
Phosphorylation at Thr308 (by PDK1) and Ser473 is required for full activation.
AKT promotes cell survival by inhibiting apoptosis through phosphorylation of Bad, Foxo1, c-Raf, and caspase-9.
Deregulation of AKT is implicated in many cancers and negatively regulated by PTEN and LY294002.

Specificity Verification: ELISA, flow cytometry, and western blotting.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated for intracellular staining of normal human peripheral blood cells.
Use at 5 µL (0.5 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.


Staining Protocols

Protocol A (Two-step): For cytoplasmic proteins; offers flexibility for surface and intracellular detection.
Protocol B (One-step): For nuclear proteins; suitable for transcription factors and phosphorylated proteins.
Protocol C (Two-step Fixation/Methanol): Best for phospho-specific signaling discrimination; limited surface staining.
