Target Information

CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa depending on phosphorylation.
It is expressed on mature and most malignant B cells, a subset of T lymphocytes, and follicular dendritic cells.
CD20 regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation.
It associates with lipid rafts, with intensity depending on extracellular triggering and BCR aggregation.
After receptor ligation, CD20 colocalizes with BCR but remains on the cell surface after BCR endocytosis.
CD20 is a key target for antibody-mediated B-cell depletion therapies and is highly expressed in most B-cell malignancies.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
