Target Information
Estrogen Receptor beta (ER beta, NR3A2) is approximately 55 kDa, with multiple isoforms (36–59 kDa). Initially cloned from human testis, ER beta belongs to the steroid/thyroid hormone receptor superfamily. It functions as a ligand-activated transcription factor that binds estrogen, undergoes conformational change, dimerizes, and interacts with estrogen response elements (ERE) in DNA to regulate gene transcription.
Two forms exist—ER alpha (ESR1) and ER beta (ESR2)—with similar estrogen binding affinities. Some ER beta isoforms inhibit ER alpha activity in reproductive tissues. ER beta is broadly expressed and may act as a tumor suppressor, relevant to cancers, autism spectrum disorder, medullary thyroid carcinoma, De Quervain Disease, and cardiovascular disease risk.
