Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa depending on phosphorylation. It is expressed on mature and most malignant B cells, as well as on a subset of T lymphocytes and follicular dendritic cells. CD20 expression is synchronous with surface IgM and regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation. It associates with lipid rafts, with the intensity of association depending on extracellular triggers and BCR aggregation. CD20 remains on the cell surface after receptor ligation and is a key target for antibody-mediated B-cell depletion therapies. Dysfunction of CD20 is linked to Ms4a1-related common variable immune deficiency.
