Product Specific Information

Description:
The H57-597 monoclonal antibody reacts with the beta chain of mouse TCR. TCR beta is expressed by thymocytes in a developmentally regulated manner and by a majority of peripheral T cells. Crosslinking of the TCR complex with H57-597 induces activation and proliferation of T cells or apoptosis depending on assay conditions. It is used as a phenotypic marker for TCR beta expressing T cells.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested with mouse thymocytes and splenocytes at ≤0.8 µg/test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Antibody titration recommended for optimal performance.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.

Buffer Information:
This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance. Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton™ technology with engineered nucleic acid dye structures for optimized fluorescent signatures and reduced spillover/spread.

Target Information

T cell receptors (TCRs) are essential for discriminating foreign from self-peptides presented by MHC class II molecules, enabling adaptive immune responses.
Self-peptide recognition is associated with autoimmune diseases, while mutant self-peptides are linked to tumors.
TCR engagement by bacterial superantigens can cause toxic shock syndrome.
Autoantibodies to V beta segments of TCRs have been found in RA and SLE patients, potentially modulating inflammatory responses.
Most human T cells express TCR alpha-beta with CD4 or CD8, while gamma-delta T cells reside mainly in epithelial tissues and exhibit distinct antigen recognition and immunoregulatory activity.
