Product Specific Information
For reconstitution, add 100 µL of distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).
Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or metabolic stress. p53 upregulates stress response genes such as p21waf1, leading to G1 or G2/M arrest.
The structure of p53 includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerization domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is crucial for p53 activation and stabilization. Ser392 phosphorylation is associated with tumor formation. p53 plays roles in aging, oxidative stress, and learning and memory processes. It is typically expressed at low levels in normal cells but highly expressed in transformed cells, contributing to malignancy. Mutations in TP53 lead to loss of tumor suppressor function and are frequently observed in human cancers, including germline mutations in Li-Fraumeni syndrome.
